The transcription factors Fra-1, c-Fos and c-Jun modulate the activity of phospholipases PLA2, PLC and Sphingomyelinase

The Immediate Early (IE) oncoproteins Fra-1, cFos and c-Jun are transcription factors involved in normal cell development and differentiation and linked with tumor growth when deregulated. c-Fos and Fra-1 interact with endoplasmic reticulum membranes both in vivo and in vitro (1).
Biophysical studies define these proteins as amphitropic, flexible and Intrinsically Unfolded. Comparative studies of the molecular interfacial behavior of Fra-1, c-Fos and c-Jun with techniques such as Langmuir compression and Gibbs adsorption isotherms, and epifluorescence microscopy, show that the surface properties of the three are overall similar, whereas they can interact with membrane phospholipids in a distinguishable manner (2).
We explored the ability of Fra-1, c-Fos and c-Jun to modulate phospholipid degradation by phospholipases (PLA2, PLC and sphingomyelinase). A comparison of the modulation of phospholipase activity by Fra-1 and c-Jun with that of c-Fos on the same enzymes (3) allows to envisage a complex combination of causes to the overall effect.
