Research in the field of theragnosis includes the development of pharmaceutical formulations which combines therapeutic and diagnostic properties. In this context, nanotechnology has offered new opportunities for the development of smart theranostic platforms owing to its outstanding properties. Thereby, the integration of targeting, imaging, and therapeutic properties in a single nanosystem results in promising “nanotheranostics”.Particularly, in stimuli-responsive nanotheranostics the therapeutic effect or the detection property takes place in response to an environmental or external stimulus pH, redox potential, temperature, magnetic field, light, or specific biomolecules. The next generation of smart nanotheranostics should be capable of tune pharmacokinetics and biodistribution, as well as achieve their functions of treatment and noninvasive monitoring on the specific pathophysiological site.In this chapter, we summarize the most relevant physicochemical aspects of novel stimuli-responsive nanotheranostics. Recent examples of functional clinically relevant nanotheranostics are addressed here under subheadings according to the inorganic material involved.
