Profiling of the IL-17 family cytokines and receptors expression on different murine tumors and their impact on tumor progression

IL-17 cytokines play important roles in protecting the host against extracellular pathogens and promoting inflammatory pathology in autoimmune diseases. In cancer, the role of IL-17 is still controversial and both pro and antitumoral role have been attributed. Previously, our team and others demonstrated that IL-17 family plays a central role in the induction of CD8+ T cell (CTL) and NK responses. Among the protumoral effects IL-17 could act as a growth factor signaling directly on tumor cells. IL-17A and IL-17F signal through a receptor complex formed by two units of IL-17RA and one unit of IL-17RC. Also, IL-17RA could form a receptor complex with IL-17RD to signalize IL-17A. Therefore, we evaluated the transcripts levels of IL-17A and IL-17F cytokines and of IL-17RA, IL-17RC and IL-17RD receptors of the IL-17 family in tumor cell lines maintained in vitro. We evaluated B16-SIY (melanoma), MCA-OVA, MC57-SIY (fibrosarcoma), EL4-SIY (lymphoma) and C1498-SIY (acute myeloid leukemia) murine tumor cell lines. We determined that the cell lines analyzed expressed IL-17RA, IL-17RD and IL-17F transcripts. Among them, interestingly, B16-SIY showed the highest expression of IL-17RA (p<0.01) and of IL-17RD (p<0.001) transcripts level. IL-17RC transcripts were expressed in MCA-OVA, B16-SIY and MC57-SIY tumor cell lines, among them B16-SIY also had the highest levels of transcripts. On the contrary, IL-17RC transcripts were not expressed in neither EL4-SIY nor C1498-SIY tumor cell lines, indicating that in this cell lines the conventional receptor complex RA-RC is not available. Moreover, C1498-SIY and B16-SIY tumor cells expressed the highest level of IL-17F transcripts (p< 0.001 and p<0.05, respectively). EL4-SIY was the only cell line that expressed IL-17A, while the rest of the tumor cells evaluated lacked expression of this transcript.
