at spleen DC and bone marrow-derived DC wereisolated and characterized by morphology and flowcytometry. We found a CD8a1 DC subpopulation representing19?48% (27.4 6 12.0) of total spleen DC. TheOX-62 expression on total spleen DC was 41?59%(51.8 6 7.5). Myeloid bone marrow-derived DC werenegative for CD8a and OX-62. We demonstrated thecoexpression of CD8a and OX-62 molecules, at least ina portion CD8a1 spleen DC. Both CD8a1 and CD8a2spleen DC subpopulations separated by MACS wereable to induce an in vivo primary immune response toOVA. The immune response induced by the CD8a2 DCsubpopulation was higher (P < 0.05). We identified aCD8a1 DC subpopulation in rat spleen less effective ininducing an immune response than CD8a2 DC. Moreover,our results suggest the presence of DC subpopulationswith different lineages in DC preparationsbased on OX-62 expression.
