Resumen:
ABSTRACTTrophoblastcelldifferentiationintomigratoryextravilloustrophoblasts(EVT)iscriticalforhumanplacentationandembryonicdevelopment.KLF6isazinc-fingertranscriptionfactorthathasacellularandsignaldependenttumorsuppressoractivity.klf6knockoutmicediein-uterowithaphenotypeincludingimpairedplacentaldevelopment,howeveritsroleinplacentaispartiallyknown.WehavedemonstratedthatKLF6isrequiredforpropervilloustrophoblastdifferentiationanditsexpressionisregulatedbyhypoxia.InordertoevaluatewhetheritmodulatesEVTmigrationweperformedwound-healingandwesternblotanalysisinKLF6silencedHTR8/SVneocells(humanfirsttrimestertrophoblastswithanEVTphenotype).Confluentcellmonolayers,transfectedwithaKLF6-specificsiRNA(siK)orcontrolsiRNA,were?scratched?andcellmigrationandproteinexpressionwereassessedafter4and24h.CellmigrationtendstobehigherinsiKcellscomparedtocontrol,althoughdifferencewasstatisticallynon-significant.Proteinexpressionlevelsofβ-catenin,connexin-43,α5andβ1integrinsubunitsremainedessentiallyunmodified.PresentresultssuggestthatdownregulationofKLF6doesnotaltertheEVTdifferentiationmarkersanalyzed.FurtherstudiesarerequiredtodeterminethepossibleroleofKLF6inEVTmigration.SupportedbyFONCyT,CONICET,SECyT-UNC
