Novel ionic hyaluronic acid-vancomycin complexes for inhalation

Polyelectrolyte-Drug complexes are interesting alternatives to improve drug performance. Vancomycin (VAN) is an antimicrobial used to treat lung infections caused by MRSA in patients with cystic fibrosis. However, nowadays there are not efficient and secure inhalable formulations containing VAN. This work is focused in the preparation and characterization of novel ionic complexes between hyaluronic acid (HA) ? a biocompatible bioadhesive polymer ? and VAN. HA-VAN complexes were easily prepared in aqueous dispersion loading different proportions of VAN. Glucose and NaOH were used to achive isosmolarity and pH approximate to 7, respectively. The ionic interaction between HA and VAN, in freeze-dried solid state, was evidenced using FT-IR and thermal analysis. In contact with water, the complexes were easily redispersed. Dispersions contaning ≤50% of VAN were physically stable at least for a week. They exhibited negative Z potential (-16,5; -23 mV), pseudoplastic flow behavior and were nebulized appropriately. Microbiology assay of complexes showed similar antimicrobial activity than VAN solutions. Release profiles of HA-VAN showed extended release of VAN with anomalous non-fickian transport. These preliminary results could be useful for the elaboration of inhaled VAN with superior properties in comparison with those currently available.
