he present study independently evaluates safety, antigen-specific humoral immunity and neutralizing antibodies response to Sputnik V vaccine. Since SARS- CoV- 2 lineage P.1 have spread rapidly in Argentina and given that neutralization of P.1 by Sputnik vaccine-elicited sera has not been issued yet, we also evaluated the efficacy of Sputnik-immunized response faced to P.1. We corroborate the safety of the vaccine since no severe adverse event was detected. Vaccine efficacy, measured by disease infection rate within 4 months was 97.61%. We find that Sputnik V induces strong humoral response in immunized individuals since 99.65% had detectable antibodies after receiving the second dose (at day 42); however 23.15% had no detectable Neutralizing Antibodies. Regarding activity against P.1, our results showed that, although a slightly reduction in Neutralization ability, P.1 variant does not escape the immunity induced by Sputnik vaccine neither to natural immunity in previously infected by
