During aging, T and B cells change their capacities to react against antigens. No data is available whether DCs could participate on these changes. Here, we analyzed the phenotypic composition and cytokine secretion profile of DCs from aged (18-month old) Balb/c mice. In spleen of aged mice, the proportion and number of CD11c+ cells is reduced 30%, compared to their counterparts in young (3-month old) mice. The proportion of B220+ DCs was increased in spleen of aged mice, whereas no difference was observed in the proportion of CD8a+ or CD4+ DCs. In naïve conditions, no differences in CD86 expression were observed, but DCs from aged mice expressed lower levels of I-Ad molecules. Purified spleen DCs from aged mice, incubated overnight with CpG ODNs (CpG motif-containing oligodeoxynucleotides) or with LPS produced more IL-10 and less IL-12p40 than DCs from young mice. In contrast, we found that in mesenteric lymph nodes (MLN) from aged mice, DCs doubled the percentage observed in MLN form young mice. No phenotypic differences were observed in MLN from aged mice. Additionally, bone marrow (BM) cells from aged mice could generate BM-derived DCs (BMDCs) as effectively as BM cells from young mice. The proportion of CD11c+ cells in BMDCs from aged mice, generated either in the presence of GMCSF or Flt3-L, was similar to young counterparts. BMDCs from both ages could equally maturate upon stimulation with CpG ODNs. Splenic DCs from aged mice show some changes that could affect the innate and adaptative immune response in spleen.
