Cellular Immune and Histopathological Events During Experimental Autoimmune Prostatitis (EAP) Development

To investigate the time course of cellular immune and histopathological events Wistar rats were immunized with male accesory glands homogenate (MAG) and Complete Freund´s Adjuvant (CFA) on day 0 and 21. CFA inoculated and non-treated rats served as controls. Delayed hipersensitivity (DTH), migration ihibitory factor (MIF) and antigen-specific T cell proliferation test were realized on day 0, 14, 21 and 28 after first immunization. Histological findings were evaluated in the prstate gland at the same time. T cell subsets, macrophages infiltrating cells and Class II MHC antigens (IA and IE) expresión by epithelial cells were analysed by IF. In EAP rats, DTH response were positive at 7, 14, and 28 days adn significantly higher than the results observed in treated and non-treated control rats. The MIF test was positive in approximately 33% of animals in all periods analizad. Proliferative responses were positive in 33 percent of the cases at day 7, 25 percent at 14 and 21 days, increasing of 50 percento of the cases at day 28. The lesion in the prostate gland was evaluated counting the number of infiltrating cells/mm2. At 7 days infiltrating cell values (42,9±29,9) in EAP rats were similar to treated (30,4±18,3) and non-treated rats (40,2±7,3). The number of mononuclear cells increased in EAP rats at 14 days (117±87,5). These values of inflammatory cells were maintained, with slight variations, at 21 days (98,9±39,6)and 28 days (151±85,3) but not reaching significant differences. In several cases this infalmmatory foci was accompanied by fibrosis and flatening of glandular epithelium. CD4 and CD8 positive cells were observed, in similar relation in all periods studied. Macrophages mAb showed isolated positive cells in the glandular interstitium. Ia epuithelial expresión was occasionally observed in the time course study, in contrast IE positive epithelial cells was evident in all periods analyzed except on day 14, when a low intensity was observed. The time course results of the immunopathological events adn the previously demosntrated low numbers of Ig-secreting cells, suggest the importante of the cellular immune response in the development of chronic prostatitis. Yhe different expresión of IA and IE antigens with the presence of infiltrating lymphocytes could mediate the cellular immune responseduring the development of the pathology.
