Resumen:
Metabolic Syndrome (MetSyn) is a complexdisorder of metabolism that affects differentorgans including the retina, triggering retinopathies.These diseases present, in additionto vascular alterations, a progressive neuronaldegeneration. In this sense, α2-macroglobulin(α2-M) has been proposed as a mediator of neurotoxicity.Considering that the neurodegenerationprecedes microvascular lesions in retinopathies,in this work, it was proposed to evaluategene and protein expression of α2-M in a mousemodel of MetSyn that exhibits signs of non-proliferativeretinopathy, such as neuronal impairmentand mild vascular alterations. C57BL/6(WT) and apolipoprotein E knockout (ApoEKO) mice fed with normal diet (ND) or 10%w/v fructose diet (FD) in drinking water from 2months of age were used. After 4 months of dietretinal extracts were obtained for Western blotand qRT-PCR analysis. Data was statisticallyanalyzed by two-way ANOVA and Bonferronipost hoc (p ≤ 0.05). All experimental procedureswill be carried out following the standardsof The Association for Research in Vision andOphthalmology (ARVO) and the ethics committeeof Chemical Sciences Faculty. The resultsshowed α2-M mRNA expression in mice retinaand a significant increase of α2-M proteinexpression in retinal extracts of ApoE KO micewith both diets, thus α2-M is synthetized in themouse retina and their expression is increasedin mice with metabolic alterations. These resultsare more than encouraging and strongly supportthe initiation of studies that allow us to knowthe role of α2-M in early stages of retinopathyinduced by MetSyn.
