Determination of Drug Loading Efficiency and Drug Release of Loperamide from Polymeric Nanocarriers.

Resumen:
Loperamide was selected as model drug. Nanocarriers were fabricated by nanoprecipitation of loperamide in a polymethacrylatederivative by adding an aqueous solution containing poloxamer 407 (P407) 0.5% w/v into an acetone-based organic phase, containingthe polymer and the model drug. Loperamide nanocrystals were obtained by the nanoprecipitation of loperamide/methanol+acetone inwater. Size, polydispersity index (PdI), and zeta potential (ZP) were determined in a Malvern Zetasizer Nano ZS. Loading efficiencywas determined by centrifugation and diafiltration. Drug release kinetics was determined by dialysis and diafiltration. Loperamide wasquantified in a UV-Vis spectrophotometer or a RP-HPLC/UV-Vis.
