After the oral administration of the polysaccharide chitosan discrete changes occur at the inductive sites of the mucosal immune system that include cell recruitment and subpopulation redistribution. The intestinal epithelium participates actively in response to luminal stimuli through the expression of pro-inflammatory genes and the secretion of cytokines and chemokines. Our purpose was to characterize events triggered by chitosan at the epithelial and inductive sites. We fed rats 200 µl of acetic acid (diluent group) or 1, 3 or 5 mg of chitosan and 16 h later we purified epithelial, Peyer´s patches (PP) and mesenteric lymph node mononuclear (MNL) cells and we obtained total RNA. The expression of TNF-a (p<0.05) and MCP-1 (p<0.05), but not IL-12 nor IL-8 mRNA (p=NS) was up-regulated in a dose dependent manner after the feeding (RT-PCR). In PP a significant increment in IL-4 (p<0.05), diminished TNF-a (p<0.05) and unmodified IL-12 mRNA expression was observed. In MLN, chitosan administration induced increments in the percentage of OX62+ and IL-10+ cells. Also a trend in the increment of OX62+IL-10+ cells was observed. Chitosan up-regulates the expression of chemokines and cytokines that participate in cell recruitment while settles a non-inflammatory environment at the inductive sites.
